Column chromatographic purification and monosaccharide composition of polysaccharide extracted from Agaricus bisporus stipe

Abstract
Using Agaricus bisporus stipe as material,firstly,the A. bisporus stipe polysaccharide was extracted by ultrasonic extraction,and purified using column chromatography. Then the purity of polysaccharides isolated by column chromatography was examined by UV spectrum and column chromatography. The infrared spectrum and monosaccharide composition of the isolated polysaccharides were also analyzed. Results showed that the extracted polysaccharide was divided into 4 components by DEAE Sephadex A-25 purification,and the components eluted by distilled water and 0.1 mol/L NaCl were collected. UV spectrum scan showed that there were no nucleic acids and proteins in both the elutes,and they were also showed a single peak using Sephadex G-200. Infrared spectrum showed the both components contained characteristic absorption peaks of polysaccharide,and they were β-pyranose polysaccharides. The polysaccharide eluted by distilled water was composed by glucose,galactose,mannose,xylose and arabinose. And the 0.1 mol/L NaCl eluting component consists of glucose,galactose,mannose,xylose and arabinose,rhamnose,glucosamine and galactosamine.
