Research of construction of DNA fingerprinting for malt varieties

  • Malt as the main raw materid of beer,assuring varietal autherticity in malt is very important for quality control in blewing. As the second generation of malecular makers,microsatellites are widely used in plant species diversity and warietry indertification. To dovelop a DNA fingerprinting of malt varieties based on simple sequence repeats(SSR),six SSR primer pairs with high polymorphisms were successfully screened out from 105 candidates,and 5.83 genotypes were detected by each SSR primer pair on an average with the range from 3 to 10. Therefore,combination of 6 pairs of primers could effectively distinguish 21 kinds of malt varieties. Furthermor affer analysis of alleles,four central primer pairs with high polymorphisms could distinguish 21 varieties of malt using specific alleles,This method anabled us to establish the DNA fingerprinting of the corresponding malt varieties and provide technical support for the malt business of breweries.
