Screening and identification of a strain producing polyhydroxybutyrate

A polyhydroxybutyrate( PHB) strain was isolated from sewage by Nile blue fluorescence staining combining with Sudan black B,which was based on glycerol as the carbon source. A series of analyses on physiological and biochemical characterization and sequence analysis of 16 S rRNA revealed that it belonged to the genus Pseudomonas pudita,which named Pseudomonas pudita PP25.PHB was extracted with sodium hypochlorite- chloroform.Finally,a high yield PHB strain named PP25 was determined by UV spectroscopy,Fourier transform infrared spectroscopy and nuclear magnetic resonance spectroscopy,yield of PHB was 2.456 g / L.
