Abstract
The culture processes and culture solution components of γ-aminobutyric acid( GABA) accumulation in potato using hypoxia aerating method were researched in this paper. Firstly,the culture processes of GABA accumulation in potato was optimized using orthogonal experimental design. Subsequently,the optimal culture solution components on GABA accumulation of potato were investigated using Box-Behnken design. The results showed that the optimum culture processes of GABA accumulation in potato under hypoxia aerating were culture time 4h,temperature 20℃,and p H of culture solutions 5.8.Range analysis indicated that p H of culture solutions was the most important influence factor,followed by culture temperature,and culture time was the last. Under these conditions,the accumulation of GABA in potato was 0.3160 mg/g,which was 3.45 times of material. Box-Behnken experimental design results displayed the optimum compositions of culture solution were MSG concentration15.43 mg/m L,Ca Cl2 concentration 2.81 mmol/L,and VB6 concentration 0.03 mg/m L. Under these conditions,the highest GABA content of potato was 0.5749 mg/g,6.28-fold of material. It indicated that the optimized culture solution compositions significantly increased GABA content of potato. Analysis of variance indicated that the regression model was significant,could forecast the changes of GABA accumulation in potato excellently.
