Protective effects of torularhodin on oxidative injured PC12 cell

Abstract
In order to study the protective effects of torularhodin on oxidative injured PC12 cell. PC12 cells were randomly divided into seven groups: control group and model group ( 200μmol / L H2O2) , lycopene group ( positive control group) and treatment groupsⅠ- Ⅳ were added with torularhodin ( 1, 2, 3, 4μmol / L) under oxidative stress, respectively.The change of cell viability and cytotoxicity was observed by MTT assay and lactic dehydrogenase kit; the activities of antioxidant enzymes and malondialdehyde ( MDA) production were measured by enzyme linked immunosorbent assay; the accumulation of reactive oxygen species ( ROS) was detected by flow cytometric analysis.Results showed that in treatment groups Ⅰ- Ⅳ cell viability ( p < 0.01) was obviously increased and cell cytotoxicity ( p < 0.05) was decreased, and the activities of antioxidant enzymes ( p < 0.05) were significantly improved and the content of MDA ( p < 0.05) was reduced, and the accumulation of ROS was inhibited. What's more, treatment group Ⅲ had the best antioxidant effect. Torularhodin had protective effects on oxidative injured PC12 cell.While torularhodin was graded to four concentrations, 3μmol / L torularhodin achieved a better ability to resist oxidative stress.
