柠檬草精油的抑菌稳定性和抗氧化活性

Studies on Antibacterial Stability and Antioxidant Activity of Essential Oils from Cymbopogon citratus

  • 摘要: 目的:研究柠檬草精油的化学成分和抗氧化活性,并讨探加工储藏条件对其抑菌稳定性的影响。方法:采用水蒸气蒸馏法提取精油,气相色谱-质谱法(GC-MS)分析成分,滤纸片法测定精油的抑菌活性以及温度、紫外线、pH和CaCl2对抑菌活性的影响,采用ABTS+·、DPPH·、O2−·清除率和Fe3+还原力评估抗氧化活性。结果:柠檬草精油中共鉴定出49种化合物,含量大于1%的有12种,占总成分的88.11%,主要成分为香叶醛(26.73%)、橙花醛(18.57%)和β-月桂烯(9.30%)。柠檬草精油对3种常见腐败菌均有较强的抑制作用,对金黄色葡萄球菌的抑制圈直径(25.43±0.24 mm)显著(P<0.05)高于大肠杆菌(23.54±0.67 mm)和枯草芽孢杆菌(18.33±0.03 mm)。柠檬草精油具有良好的紫外稳定性,温度和pH对精油的抑菌效果影响较大,低温和酸性条件下其抑菌活性较强,CaCl2可增加柠檬草精油的抑菌效果。柠檬草精油具有良好的抗氧化活性,对ABTS+·、DPPH·、O2-·的清除能力和Fe3+的还原能力随着精油浓度升高显著增强(P<0.05),对ABTS+·、DPPH·、O2-·的半抑制浓度(half maximal inhibitory concentration,IC50)分别为4.36、7.09、3.13 mg/mL,是抗坏血酸的6.92、10.58和5.05倍。结论:柠檬草精油具有良好的抑菌活性和抗氧化活性,在储藏和加工过程中应避免高温和碱性条件。

     

    Abstract: Objective: To study the chemical components and antioxidant activity of Cymbopogon citratus essential oil and to and to investigate the effect of processing and storage conditions on its antibacterial stability. Methods: The essential oil was extracted by steam distillation, and its chemical components were analyzed by GC-MS. The effects of temperature, ultraviolet rays, pH, and CaCl2 on the antibacterial activity were determined by filter-paper method and the antioxidant activity was determined by the ABTS+·,DPPH· and O2· radical-scavenging assay and the reducing capacity for Fe3+. Results: A total of 49 compounds were identified, and 12 compounds with contents greater than 1%, accounting for 88.11%, with citral (26.73%), neral (18.57%), and β-myrcene (9.30%) as the main components. The essential oil of Cymbopogon citratus showed strong antibacterial activity. The inhibitory effect against Staphylococcus aureus (25.43±0.24 mm) was better than that of Escherichia coli (23.54±0.67 mm) and Bacillus subtilis (18.33±0.03 mm) (P<0.05). The antibacterial activity of the essential oil were stable under ultraviolet rays, but were unstable under different temperature and pH. The antibacterial activity was stronger at low temperatures and acidic conditions, and CaCl2 could increases the antibacterial effect. The essential oil showed good antioxidant activity, the scavenging ability of ABTS+·, DPPH·, O2−· and the reducing ability of Fe3+ increased with the concentration of essential oil(P<0.05), the IC50 of ABTS+·, DPPH·, O2−· were 4.36, 7.09, 3.13 mg/mL, respectively, which were 6.92, 10.58 and 5.05 times of ascorbic acid. Conclusion: The essential oil of Cymbopogon citratus has good antibacterial activity and antioxidant activity, high temperatures and alkaline conditions should be avoided during storage and processing.
