山楂、乌梅、葡萄皮、枸杞、桑葚复方组合物的体内降血脂作用

In Vivo Hypolipidemic Effect of the Compound Composition of Crataegus, Ume, Grape Skin, Wolfberry and Mulberry

  • 摘要: 探究山楂、乌梅、葡萄皮、枸杞、桑葚组成的复方组合物的降血脂功能。SPF级健康雄性SD大鼠50只,10只大鼠给予维持饲料作为空白对照组,40只给予模型饲料作为模型组。模型组给予模型饲料2周后,根据总胆固醇(total cholesterol,TC)水平将模型组40只动物随机分成4组,即高、中、低剂量组(1.5、0.5、0.25 g/kg·BW)和模型对照组。分组后3个剂量组每天按照10 mL/kg·BW经口给予受试样品,空白对照组和模型对照组同时给予同体积的蒸馏水。连续灌胃30 d后,测定血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白胆固醇(high density lipid-cholesterol,HDL-C)、低密度脂蛋白胆固醇(low density lipid-cholesterol,LDL-C)水平,测定肝脏系数、肝脂肪酶(hepatic lipase,HL)、脂蛋白脂肪酶(lipoprotein lipase,LPL)活性水平,通过HE染色观察肝组织脂肪变性程度。结果表明:与模型对照组相比,高、中剂量组大鼠血清TC、TG、LDL-C水平均显著降低(P<0.05),HDL-C水平极显著提高(P<0.01),高剂量组大鼠HL、LPL活性极显著提高(P<0.01),高、中剂量组脂滴空泡明显减少、脂滴泡缩小,因此认为该组合物具有调节血脂的作用,提示可以将其开发为新型降脂保健食品。

     

    Abstract: To explore the in vivo hypolipidemic effect of the compound composition of crataegus, ume, grape skin, wolfberry and mulberry. There were 50 SPF-grade healthy male SD rats, 10 rats were given maintenance feed as a blank control group, and 40 rats were given model feed as a model group. After the model group was given the model feed for 2 weeks, the 40 animals in the model group were randomly divided into 4 groups according to the TC level, namely high, medium and low dose groups (1.5, 0.5, 0.25g/kg·BW) and the model control group. After grouping, the three dose groups were given the test sample orally at 10mL/kg·BW every day, and the blank control group and the model control group were given the same volume of distilled water at the same time. After continuous intragastric administration for 30 days, serum total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) were measured. The liver coefficient, hepatic lipase (HL), and lipoprotein lipase (LPL) activity levels were measured, and the degree of hepatic steatosis was observed by HE staining. The results showed that compared with the model control group, the levels of serum TC, TG and LDL-C in the high-dose and middle-dose groups significantly decreased (P<0.05), and the HDL-C level significantly increased (P<0.01). The activities of HL and LPL in rats significantly increased (P<0.01), and lipid droplet vacuoles and lipid droplet bubbles were significantly reduced in high and medium dose groups. Accordingly, it is believed that the composition has the effect of regulating blood lipids, indicating that it could be developed as a new type of fat-lowering health food.
