辐照处理对桦褐孔菌多糖抗疲劳作用的影响

Effect of Irradiated Process on Anti-fatigue Activity of Inonotus obliquus Polysaccharide

  • 摘要: 目的:研究经过辐照处理的桦褐孔菌多糖对小鼠的抗疲劳作用。方法:用60Co-γ射线进行辐照,得到5、10、20、30 kGy辐照剂量和未辐照的桦褐孔菌菌粉制得多糖。分为对照组、辐照高中低剂量组和未辐照高剂量组,对雄性昆明种小鼠进行四周连续灌胃,测定小鼠爬杆、力竭游泳时间,肝糖原(LG)、肌糖原(MG)、全血乳酸(BLA)含量、乳酸脱氢酶(LDH)活性、血清尿素氮含量(SUN)、骨骼肌中超氧化物歧化酶(SOD)活性和丙二醛 ( MDA)含量的变化。结果:实验测得的最优辐照剂量为20 kGy。各剂量组对小鼠体重没有显著影响(P>0.05),辐照高、中剂量组与未辐照组显著延长了小鼠爬杆时间、力竭游泳时间(P<0.05)。辐照桦褐孔菌多糖增加了小鼠肝糖原、肌糖原含量,最高为28.90、10.58 mg/g,减少了小鼠BLA、SUN及MDA含量,增加了小鼠LDH、SOD活性,最高为5308.80 U/L、11.44 nU/mg。且辐照高剂量组LG、MG、LDH、SUN、SOD、MDA与未辐照组相比均差异显著(P<0.05),两组BLA差异不显著(P>0.05)。结论:该实验由此证明辐照处理桦褐孔菌多糖与未辐照组相比抗疲劳作用有进一步提高,且辐照处理桦褐孔菌多糖能够在一定程度上缓解疲劳症状。

     

    Abstract: Objective: To study the anti-fatigue effect of irradiated Inonotus obliquus polysaccharide on mice. Method: 60Co-γ rays were irradiated to obtain 5, 10, 20, 30 kGy irradiation doses and unirradiated Inonotus obliquus bacteria powder to produce polysaccharides. Divided into a control group, an irradiated high-medium-low-dose group, and a non-irradiated high-dose group. Male Kunming mice were given four weeks continuous gavage to determine the rod climbing, exhausted swimming time, changes in liver glycogen(LG), muscle glycogen(MG), blood lactate(BLA), lactate dehydrogenase(LDH), serum urea nitrogen(SUN), superoxide dismutase(SOD) and malondialdehyde(MDA) in skeletal muscle. Result: The optimal radiation dose measured in the experiment was 20 kGy. Each dose group had no significant effect on the body weight of the mice(P>0.05). The high and medium dose groups and the non-irradiated groups significantly prolonged the rod climbing time and exhausted swimming time of the mice(P<0.05). Irradiation of Inonotus obliquus polysaccharides increased the contents of liver glycogen and muscle glycogen in mice to the highest levels of 28.90 mg/g and 10.58 mg/g, reduced the contents of BLA, SUN and MDA in mice, and increased LDH and SOD activities in mice. The highest activity was 5308.80 U/L and 11.44 nU/mg. Moreover, LG, MG, LDH, SUN, SOD, MDA in the high-dose irradiated group were significantly different from those in the non-irradiated group(P<0.05), and there was no significant difference in BLA between the two groups(P>0.05). Conclusion: This test proves that the anti-fatigue effect of the irradiated Inonotus obliquus polysaccharide is further improved compared with the non-irradiated group, and that the irradiated Inonotus obliquus polysaccharide can relieve fatigue symptoms to a certain extent.
