Abstract:
Objective: Using the branches of
Actinidia argutaas the raw material to optimize the extraction technology of Total Triterpenes from the branches of
Actinidia arguta by response surface methodology (RSM). The
in vitro anti-inflammatory activity of AABTT was analyzed. Methods: 5% Vanillin-glacial acetic acid-perchloric acid method was used to quantitatively analyze AABTT. On the basis of single factor experiment, response surface methodology was applied to optimize the main extraction conditions. Using hyaluronidase activity and the inhibition of serum albumin denaturation as an anti-inflammatory evaluation index to evaluate the anti-inflammatory activity of AABTT. Results: The best conditions for the response surface optimization were as follows: ethanol concentration 77%, liquid-material ratio 40:1 mL/g, extracting time 40 minutes, and ultrasonic power 400 W. The extraction content of Total Triterpenes was (58.42±0.36) mg/g under the optimal conditions. The anti-inflammatory activity of AABTT (4 mg/mL) was 81.48%±0.48% for the hyaluronidase and 71.09%±0.39% for the denaturation of bovine serum albumin. Conclusion: The extraction conditions of the AABTT are optimized by response surface method, and it was preliminarily showed that the AABTT had significant anti-inflammatory activity.
