黄芪对2型糖尿病大鼠碱性磷酸酶与炎症反应的影响

Effect of Astragalus on Intestinal Alkaline Phosphatase and Inflammatory Response in Type 2 Diabetic Rats

  • 摘要: 目的:探究黄芪对2型糖尿病大鼠碱性磷酸酶、细菌内毒素和血清中炎症因子的影响。方法:采用高糖高脂饮食+链脲佐菌素腹腔注射建立2型糖尿病模型,连续给予大鼠灌胃黄芪水溶液(0.25、0.5 g/kg),测定大鼠体质量及血糖变化、粪便中肠道碱性磷酸酶(IAP)的活力及细菌内毒素(LPS)的浓度变化并用MSD多因子方法检测血清中炎症因子(白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、γ干扰素(IFN-γ)、白细胞介素-1(IL-1β)、白介素-4(IL-4)、白介素-5(IL-5)、白介素-10(IL-10)、白介素-13(IL-13)、生长调节致癌基因α(KC/GRO))浓度变化的分析。结果:与模型组相比,黄芪剂量组空腹血糖浓度显著降低(P<0.05),IAP活力显著增加(P<0.05),LPS浓度显著降低(P<0.05),且血清中促炎因子IL-6、TNF-α、IFN-γ、IL-1β的浓度显著减少(P<0.05),而抗炎因子IL-4、IL-5、IL-10、IL-13的浓度显著增加(P<0.05),趋化因子KC/GRO的浓度显著下降(P<0.05)。结论:黄芪可以促进肠碱性磷酸酶活性,并能够有效调节T2DM小鼠血糖及炎症反应。

     

    Abstract: Objective: To explore the effects of Astragalus on alkaline phosphatase, bacterial endotoxin and serum inflammatory factors in type 2 diabetes rats. Methods: A high-sugar and high-fat diet + intraperitoneal injection of streptozotocin was used to establish a type 2 diabetes model. The rats were continuously administered Astragalus aqueous solution (0.25, 0.5 g/kg) and measureed the changes in body mass, blood sugar, the activity of intestinal alkaline phosphatase (IAP) in feces and the changes in bacterial endotoxin (LPS) concentration. MSD multi-factor method was used to detect serum inflammatory factors (Interleukin-6 (IL-6), Tumor necrosis factor-α (TNF-α), Interferon-γ (IFN-γ), Interleukin-1 (IL-1β), Interleukin-4 (IL-4), Interleukin-5 (IL-5), Interleukin-10 (IL-10), interleukin-13 (IL-13) growth regulatory oncogene α (KC/GRO)) concentration changes. Results: Compared with the model group, the fasting blood glucose concentration in the Astragalus dose group was significantly reduced (P<0.05), the IAP activity was significantly increased (P<0.05), the LPS concentration was significantly reduced (P<0.05), and the concentration of serum pro-inflammatory factors IL-6, TNF-α, IFN-γ and IL-1β were significantly reduced (P<0.05), while the concentration of anti-inflammatory factors IL-4, IL-5, IL-10, and IL-13 increased significantly (P<0.05), the concentration of chemokine KC/GRO decreased significantly (P<0.05). Conclusion: Astragalus could promote the activity of intestinal alkaline phosphatase and effectively regulate blood sugar and inflammation in T2DM mice.
