Abstract:
A purification method of ellagic acid(Ea)from Kushui rose waste and its antioxidant activities were studied. Firstly,the crude alcohol extracts of rose residues were purified by macroporous adsorption resin to enrich the target component. Then,the separation was performed using high-speed count-current chromatography with a two-phase solvent system consisting of n-hexane-ethyl acetate-methanol-water(2:7:3.5:7,v/v/v/v). Ellagic acid with purity of 96.2% was achieved with the flow rate of 3.0 mL·min
-1,the speed of 850 r·min
-1,the system temperature of 30 ℃ and sample loading of 800 mg,upper phase as stationary phase and lower phase as mobile phase. Furthermore,the
in-vitro antioxidant test results showed that Ea possessed the strong antioxidant effects on the clearance of DPPH· and ABTS
+·,with IC
50 values of(2.54±0.05)and(9.62±0.17) mg·L
-1,respectively. Meanwhile,the total antioxidant capacity FRAP value was(180.85±3.16) μmol·L
-1·mg
-1. This simple technology obtained the products with high purity,which provided a theoretical basis for the integrated utilization of rose waste biomass.
