离子色谱-脉冲安培法检测食品中功能性低聚异麦芽糖

Determination of Isomaltooligosaccharide in Foods by Ion Chromatography-pulsed Amperometric Detection

  • 摘要: 目的:建立一种离子色谱-脉冲安培检测法测定食品中的异麦芽糖、异麦芽三糖、潘糖的方法。方法:样品经水溶解提取,酸沉淀蛋白后,过反相固相萃取柱净化,以赛默飞CarboPacTM PA20阴离子交换柱为分析柱,以水(A)、200 mmol/L氢氧化钠(B)为淋洗液梯度洗脱,离子色谱-脉冲安培检测器定量测定。结果:在0.8 ~16.0 mg/L范围内线性关系良好,相关系数r均大于0.999,异麦芽糖、异麦芽三糖、潘糖精密度RSD分别为3.42%、2.34%、1.11%,回收率在85.63%~105.22%之间,相对标准偏差在2.48%~4.49%之间。结论:该方法前处理简单,专一性强,结果准确度高,适用于多种食品中的低聚异麦芽糖的测定。

     

    Abstract: Objective: An ion chromatography - pulsed amperometric detection method was developed for the determination of isomaltose, isomaltotriose, panose in foods. Methods: Samples were extracted by water and the protein in sample was deposited by acid, and purified with the column of guard ⅡRP, then those sugars were separated on a CarboPac PA20 high performance anion-exchange column of ThermoFisher. The water(A), 200 mmol/L sodium hydroxide solution (B) were used as mobile phases for gradient elution, and pulsed amperometric detector was used for quantitative determination. Results: The linear relationship was good within the range of 0.8~16.0 mg/L, the correlation coefficient was more than 0.999, the RSD of precision of isomaltose、isomaltotriose and panose were 3.42%, 2.34%, 1.11%, the recovery rate was 85.63%~105.22%, the RSD was 2.48%~4.49%. Conclusion:This method demonstrated to be simple with good accuracy and specifity, which is suitable for the determination of isomaltooligosaccharide in kinds of foods.
