花旗参茶对小鼠免疫功能的影响

Effect of American Ginseng Tea on the Immune Function of Mice

  • 摘要: 目的:采用SPF级BALB/c小鼠作为实验动物,评价花旗参茶对小鼠免疫功能的影响。方法:BALB/c正常小鼠随机分为阴性对照组、花旗参茶低、中、高(0.275、0.550、1.100 g生药/kg)剂量组、西洋参饮片泡水组(0.500 g生药/kg),采用脾淋巴细胞转化、迟发型变态反应、抗体生成细胞检测、血清溶血素测定、碳廓清、腹腔巨噬细胞吞噬荧光微球和NK细胞活性测定等实验方法,观察花旗参茶对小鼠免疫功能的影响,并运用网络药理学方法分析其作用机制。结果:花旗参茶对小鼠体重增长和胸腺系数均无明显影响。与阴性对照组相比,花旗参茶中、高剂量均可显著提高小鼠细胞免疫及单核巨噬细胞清除功能,高剂量还可显著提高脾脏系数、体液免疫功能及NK细胞活性,低剂量可显著增加抗体生成细胞数。β-谷甾醇和人参皂苷Rh2等为花旗参茶主要活性物质,作用机制涉及IL-17和NOD样受体等炎症相关通路及细胞凋亡信号通路。结论:花旗参茶具有较好的提高机体免疫力的作用。

     

    Abstract: Objective:To evaluate the effects of American Ginseng tea on immune function in SPF grade of BALB/c mice. Methods:BALB/c mice were randomly divided into control group,low-dose,medium-dose and high-dose American Ginseng tea group(0.275,0.550 and 1.100 g crude drug/kg,respectively),and American Ginseng soaking water group(0.500 g crude drug/kg). Experiments of spleen lymphocyte transformation,delayed type hypersensitivity,antibody-producing cell detection,serum hemolysin determination,carbon clearance,peritoneal macrophage phagocytosis fluorescent microspheres and NK cells activity were performed to observe the effect of American Ginseng tea on immune function of mice. And network pharmacology analysis was used to explore the mechanism of American ginseng tea. Results:American Ginseng tea had no significant effect on the weight gain and thymus coefficient of mice. Compared with the control group,the medium and high dose of Ginseng tea could significantly improve the cellular immunity and mononuclear macrophage clearance in mice. The high-dose group of American Ginseng tea could significantly improve spleen coefficient,humoral immune function and NK cell activity. And the low-dose group of American Ginseng tea could significantly increase the number of antibody-producing cells. β-sitosterol and ginsenoside Rh2 were considered to the major active substances in American Ginseng tea,and the mechanism of treatment may involve IL-17,NOD-like receptor and other inflammation-related pathways as well as apoptosis signaling pathways. Conclusion:American Ginseng tea has the potential to enhance immune function.
