Abstract:
Objective:To explore the regulatory effects of the tested strains on the secretion of inflammatory cytokines in THP-1 cells induced by lipopolysaccharide(LPS),and to screen probiotics with the potential function of improving the inflammatory response. Methods:THP-1 was induced into mature macrophages by phorbol ester(PMA),then lipopolysaccharide(LPS)stimulated mature macrophages to produce inflammatory responses,so as to evaluate the improvement effect of probiotics on this inflammatory response. Besides,the gastrointestinal tolerance,adhesion,bacteriostatic ability and antimicrobial susceptibility of probiotic strains with the potential function of improving inflammatory response were evaluated. Results:
Lactobacillus plantarum SS-9,
Lactobacillus plantarum SD-H9,
Lactobacillus rhamnosus SD-L8 and
Lactobacillus reuteri WX-94 had the potential function to improve inflammatory response. SS-9 and SD-H9 could significantly reduce the secretion of IL-1
β in inflammatory model cells(
P<0.05). WX-94 and SS-9 could significantly reduce the secretion of IL-6(
P<0.05). Four kinds of strains of bacteria had high gastrointestinal tolerance,adhesion ability,and had the potential to safely pass through the intestine. Four kinds of strains of bacteria could inhibit the growth of pathogenic bacteria such as
Escherichia coli,Staphylococcus aureus,Pseudomonas aeruginosa and
Enterobacter cloacae in varying degrees,and had the potential to inhibit intestinal infection. Results of antibiotic susceptibility showed that none of the four kinds of strains had a risk of antibiotic tolerance. Conclusion:
Lactobacillus plantarum SS-9,
Lactobacillus plantarum SD-H9,
Lactobacillus rhamnosus SD-L8 and
Lactobacillus reuteri WX-94 can be developed and applied as probiotics with potential inflammatory regulation functions.
