凤凰单枞多酚提取工艺优化及其体外抗氧化和α-葡萄糖苷酶抑制活性研究

Extraction Optimization of Polyphenols from Phoenix Dan Cong and Its in vitro Antioxidant Capacity and Inhibitory Activity of α-Glucosidase

  • 摘要: 为促进特色茶资源的开发利用,本研究以潮汕地区特有的乌龙茶凤凰单枞为原料,采用超声辅助提取茶多酚,以乙醇体积分数、液料比、提取时间和温度为考察因素,在单因素实验基础上,利用Box-Benhnken实验设计对凤凰单枞茶多酚的提取工艺进行优化,并探究所得多酚提取物体外抗氧化及对α-葡萄糖苷酶的抑制活性。研究结果表明,凤凰单枞多酚提取最佳工艺条件为乙醇体积分数70%、提取温度57 ℃、提取时间34 min、液料比42:1 mL/g,多酚得率可达18.18%±0.23%。该工艺条件下提取所得凤凰单枞多酚对DPPH·和·OH具有较强的清除能力,清除能力与多酚质量浓度呈一定量效关系,其IC50分别为8.42和24.70 μg/mL,均低于相同质量浓度下的VC。此外,该多酚提取物对α-葡萄糖苷酶活性具有显著抑制作用,IC50为7.24 μg/mL,抑制效果优于对照组阿卡波糖。该研究为凤凰单枞天然抗氧化及降糖功效的利用提供理论依据。

     

    Abstract: To promote the development and utilization of regional tea resources,the polyphenols in Phoenix Dan Cong,a typical oolong tea from Chaoshan district,was extracted by ultrasound-assisted treatment. The ethanol concentration,liquid/solid ratio,extraction time and temperature were investigated by single factor test and Box-Behnken response surface methodology to acquire the optimum extraction process. Meanwhile,the antioxidant capacity and α-glucosidase inhibitory effect of extracted polyphenols were also evaluated. The results indicated that the optimum extraction parameters were as follows:Ethanol concentration 70%,extraction temperature 57 ℃,extraction time 34 min,liquid/solid ratio 42∶1 mL/g,and the extraction rate of polyphenols reaches 18.18%±0.23%. The in vitro antioxidant test showed that the extracted polyphenols had significant DPPH and OH free radical scavenging activity which were mass concentration-dependent. The IC50 values were 8.42 and 24.70 μg/mL respectively,which were lower than VC at the same mass concentration. Moreover,the extracted polyphenols exhibited strong inhibitory activity against α-glucosidase better than acarbose,with an IC50 of 7.24 μg/mL. These results provided a theoretical basis for utilization of the natural antioxidation and hypoglycemic effect of Phoenix Dan Cong.
