Abstract:
Objective:Based on metabolomics technology,the global mechanism of Fenggang Zinc-Selenium Tea on metabolic regulation of normal rat was explored. Method:Rat were orally administered with steamed water(10 mL/kg/d,as control),high dose(1 g/kg/d)and low dose(0.5 g/kg/d)of Fenggang Zinc-Selenium Tea for 6 weeks,respectively. The pathological changes of rat liver was observed. The SOD and MDA were detected in rat plasma. The metabolites in plasma were derived with BSTFA and then analyzed by gas chromatography-mass spectrometer(GC-MS),then the data were anylzed by principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)revealel the effect of Fenggang Zinc-Selenium Tea on metabolite profile in rats. The differential metabolites were screend. Metaboanalyst was used to obtain the metabolic pathway. Result:HE staining results showed that there were no obvious pathological changes in the liver of the control group and the tea treatment group. After 4 weeks,the MDA content in the high and low dose groups were significantly lower than those in the control group(
P<0.05). There was a significant difference in the metabolic profile of the tea treatment group compared with the control group. And the difference metabolites including the butyric acid,L-proline,succinic acid,inositol and stearic acid were screed. And these differential metabolites mainly involve three metabolic pathways including phosphoinositide metabolism,arginine and proline metabolism,and tricarboxylic acid cycle. Conclusion:This study preliminarily revealed the biological metabolic mechanism of Fenggang Zinc-Selenium Tea affecting the metabolic regulation of rat through metabolic pathways such as phosphoinositide metabolism,arginine and proline metabolism,and tricarboxylic acid cycle.This provides a theoretical basis for the further research and development of Fenggang Zinc-Selenium Tea.
