超声-闪式协同提取白及须根多糖工艺优化及其抗肿瘤活性

Optimization of Ultrasonic-Flash Synergistic Extraction and Antitumor Activity of Polysaccharide from Fibrous Root of Bletilla striata

  • 摘要: 以白及须根为原料,多糖提取量为考察指标,在单因素试验的基础上,选取料液比、闪提电压、超声功率和提取温度4个影响因素为自变量,采用响应面法对超声-闪式协同提取白及须根多糖的工艺参数进行优化,并通过建立皮下荷瘤小鼠模型,灌胃处理后观测白及须根多糖对S180荷瘤小鼠的抑瘤作用,测定血清中TNF-α及IL-2水平。结果表明,超声-闪式协同提取白及须根多糖的最佳工艺为:以去离子水作为提取溶剂,在料液比为1:21 g/mL、闪提电压为190 V、超声功率为82 W、提取温度为62℃的条件下提取60 s;抗肿瘤活性研究结果显示三个不同剂量组小鼠瘤重较模型组均明显降低(P<0.05,P<0.01),表明白及须根多糖对S180荷瘤小鼠肿瘤的生长具有明显的抑制作用,其机制可能是通过调节血清中TNF-α与IL-2水平来增强机体免疫功能而发挥抗肿瘤作用。

     

    Abstract: Fibrous root of Bletilla striata were treated as material and the extraction amount of polysaccharide was taken as the index. On the basis of single factor experiments,4 factors including the ratio of material to liquid,flash voltage,ultrasonic power and extraction temperature were selected as independent variables. Response surface methodology(RSM)was used to optimize the parameters of ultrasonic-flash synergistic extraction technology of polysaccharide from fibrous root of Bletilla striata. By establishing the subcutaneous tumor-bearing mice model,the anti-tumor effect of polysaccharides from fibrous root of Bletilla striata on S180 tumor-bearing mice was observed after intragastric administration,and the levels of TNF-α and IL-2 in serum were determined. The results showed that the optimum extraction process of polysaccharide from fibrous root of Bletilla striata by ultrasonic-flash synergistic extraction technology was as follows:Deionized water was used as extraction solvent and the ratio of material to liquid was 1:21 g/mL,the flash voltage was 190 V,the ultrasonic power was 82 W,the extraction temperature was 62℃ and the synergistic extraction time was 60 s. The results of anti-tumor activity showed that the tumor weight of mice in three different dosage groups was significantly lower than that of model group(P<0.05,P<0.01). The results showed that the polysaccharide from fibrous root of Bletilla striata had a significant inhibitory effect on the tumor growth on S180 tumor-bearing mice. The mechanism of anti-tumor might be that it could enhance the immune function by regulating the levels of TNF-and IL-2 in serum.
