Abstract:
The monosaccharide composition and contents of original peach gum after acid hydrolysis with trifluoroacetic acid(TFA)were discussed and determined by HPLC-ELSD. The results showed that liquid condition were:Chromatographic column Waters XBridge
TM Amide,column temperature 30℃,sample quantity 10 μL,mobile phase acetonitrile-water(84:16),and the flow rate 0.8 mL/min. ELSD conditions were:Drift tube 110℃,gas flow 2.0 L/min. The method results showed that the correlation coefficient
R2 of the standard curve in this method were between 0.9908 and 0.9999,and the concentration of solution was 0.01~2.00 mg/mL. The recovery of the samples were between 93.4% and 114.1%. And the precision,repeatability and stability were verified by experiments. The results indicated that the monosaccharides in the hydrolysates of different producing areas all included rhamnose 31.58~36.43 mg/g,xylose 37.35~48.54 mg/g,arabinose 262.04~315.13 mg/g,glucose 89.58~104.25 mg/g,galactose 305.33~358.99 mg/g. The cluster analysis of monosaccharides from different regions showed that the original peach gum from ten different producing areas were three kinds of peach gum clusters when the Euclidean distance was less than 5. Wuxi,Shengzhou and Cangnan were grouped together,Pingnan and Hekou were of one kind,Suizhou,Lijiang,and the others were together for a class.
