Abstract:
The pure conditions of puerarin was explored in Kudzu plants,and detected the preventive effect of the puerarin purification in drunkenness mice. The Kudzu’s root extract was purified under the condition of the washing times,the ethanol concentration and the ratio of solid to liquid,using glucose gel chromatographic technique. High Performance Liquid Chromatography(HPLC)was used to assay the puerarin purification,and GC was used to detect the mice model of acute intoxication of blood alcohol content. The results showed that,the optimum condition was:The volume fraction was 65% ethanol,the number of washing times was 6 times,and the ratio of solid to liquid was 4:2 mL/mg,the puerarin purification rate was 70.56%±0.85%.The testing results of HPLC were consistent with the element standard of puerarin,GC testing showed that puerarin purification group of mice had a low blood alcohol level than alcohol model group of mice,the average alcohol levels in puerarin group of mice was 3.295 mg/mL(
p<0.01),and the average alcohol levels in alcohol group of mice was 4.605 mg/mL. Glucose gel purification method for puerarin was simple and feasible,it could be used for isolation and purification of puerarin,and the puerarin purification could reduce acute intoxication of mice blood alcohol concentration,which had intervention effect to drunkenness mice.
