白灵菇蛋白质的理化及功能特性

Physicochemical and Functional Properties of Protein from Pleurotus nebrodensis

  • 摘要: 为充分开发利用白灵菇蛋白质(Pleurotus nebrodensis proteins,FPs),以残次级白灵菇为原料,采用碱浸提分段盐析法依次制备60%、80%、90%盐饱和度析出蛋白质(F60、F80、F90),统称为分段盐析蛋白质组分(FPs),测定FPs的析出率、理化性及FPs中F60的功能特性。结果表明:F60的析出率最高,达65.62%。FPs间的二硫键含量无显著性差异(p>0.05);FPs的蛋白质条带中共同含有分子量约为31 kDa的蛋白质条带,蛋白质条带间存在差异。F60较F80、F90富含分子量为14.4 kDa的蛋白质条带,具有较高的表面疏水性与荧光发射光谱峰值,β-折叠结构出现频率更高。F60具有优良的溶解性、起泡性、易消化水解,具有潜在的应用价值。白灵菇蛋白质是一种优质的新型蛋白质资源。

     

    Abstract: To further enhance the use of Pleurotus nebrodensis proteins(FPs),the exhalation rate and physicochemical properties of proteins were separated out when the percentage base saturation was equal to 60%(F60),80%(F80),90%(F90),which were named FPs collectively. And the precipitation rate,physicochemical properties of FPs and the functional properties of F60 were determined. The results showed the exhalation rate of F60(65.62%)was higher than F80 and F90. No significant difference of the content of disulfide bond was found between FPs(p>0.05). FPs had one band(at about 31 kDa)and the protein bands of FPs existed differences. F60 exhibited higher surface hydrophobicity and fluorescence intensity,and included more β-sheet than F80 and F90. F60 exhibited good solubilities,foaming capacities,and excellent in-vitro digestibilities. Pleurotus nebrodensis proteins is a kind of high grade protein resource.
