响应面法优化超声提取杏鲍菇皂苷

Optimization of Ultrasonic Extraction of Pleurotus eryngii Saponin by Response Surface Methodology

  • 摘要: 优选杏鲍菇皂苷的超声提取工艺。采用超声波辅助提取杏鲍菇皂苷,通过单因素实验,确定超声提取时间、液固比、提取温度和提取液pH对杏鲍菇皂苷得率的影响;并采用Box-Behnken响应面设计优化提取条件,建立各因素的二次回归方程模型和方差分析,确定超声辅助提取杏鲍菇皂苷的最佳提取条件为超声提取时间31 min,液固比11.5 mL·g-1,提取温度65 ℃,提取液pH8.4,此时杏鲍菇皂苷提取率可达到3.19%。优选的超声提取工艺稳定可靠,为工化业制备杏鲍菇皂苷提供了一定的理论依据。

     

    Abstract: To optimize ultrasonic extraction process of saponin from Pleurotus eryngii. In this study,the saponins were extracted from Pleurotus eryngii by ultrasonic extraction. Through single factor test,the effects of ultrasonic extraction time,liquid-solid ratio,extraction temperature and pH of extract on the yield of Pleurotus eryngii saponins were investigated. The Box-Behnken response surface design extraction experiment was used to establish the quadratic regression equation model and variance analysis of each factor. The optimal extraction conditions for ultrasonic extraction of Pleurotus eryngii saponins were obtained:Ultrasonic extraction time 31 min,liquid-solid ratio 11.5 mL·g-1,extraction temperature 65 ℃,extract pH8.4.Under this condition,the extraction rate of Pleurotus eryngii saponin was up to 3.19%.This optimized extraction process was stable and reliable,which provided a theoretical basis for the industrial preparation of Pleurotus eryngii saponin.
