微波辅助酶法优化怀山药多糖提取工艺的研究

Study on technology of polysaccharide extraction from dioscorea opposita by microwave assisted enzymatic method

  • 摘要: 建立并优化微波辅助酶法提取怀山药多糖的工艺,以期提高怀山药多糖的提取率。采用怀山药为原料,怀山药多糖提取率为指标,在微波法辅助下,加入α-淀粉酶与纤维素酶,通过单因素实验与正交实验,确定最佳提取条件为提取温度为85 ℃,提取时间为4 h,料液比1:5.5 (g/mL),微波功率为825 W。结果表明,应用微波辅助酶法,怀山药多糖提取率可达34.78%±1.19%,明显高于常规方法。

     

    Abstract: To study the technology of polysaccharide extraction from dioscorea opposite and improve the extraction yield, microwave assisted enzymatic method was established. The dioscorea opposita and the polysaccharide yield were used as materials and the index. The best extraction conditionals were optimized through the single factor experiment and orthogonal test by adding the α-amylase and cellulasel with the microwave assisted, and the optimized conditionals were the extraction temperature of 85℃, extraction time of 4 h, material to liquid ratio of 1:5.5 (g/mL), microwave power of 825 W.The results proved that the extraction yield of dioscorea opposita polysaccharide was 34.78%±1.19% by applying microwave assisted enzymatic, higher than in common methods.
