双水相法分离纯化灵芝中三萜的工艺优化

Optimization of extraction and purification process of triterpenoids from the fruitbodies of Ganoderma lucidum in aqueous two-phase system

  • 摘要: 基于亲水性醇-无机盐双水相体系,建立高效分离纯化灵芝三萜的新方法。考察亲水性有机试剂种类、无机盐(种类、浓度、pH)、样品加入量、分相后下相溶液pH对三萜得率的影响。在单因素实验的基础上,选择K3PO4溶液浓度、样品加入量、分相后下相溶液pH为实验因素,采用L9(34)正交实验优化灵芝中三萜分离纯化的最佳条件。最佳萃取条件为:10.6% K3PO4、42.5%甲醇、5%灵芝粗提液组成的双水相体系,成相后调节下相萃取液pH至 4.0,灵芝三萜提取量可达(10.13±0.19) mg/g,较碱提酸化法(7.42±0.09) mg/g提高了36.5%。甲醇/K3PO4双水相体系是一种简单、高效分离纯化灵芝三萜的新方法。

     

    Abstract: Based on hydrophilic alcohol-salt aqueous two-phase system, a rapid and efficient method for extraction and purification of triterpenoids from Ganoderma lucidum was constructed with methanol/K3PO4.The type of hydrophilic alcohol, inorganic salt (type, concentration, pH), the amount of Ganoderma lucidum extract, the pH of the lower phase was investigated by using the single factor experiment. Subsequently, orthogonal design methodology was used to explore the optimal extraction condition, the solution concentration of K3PO4, the amount of the extractive, the pH of the lower phase was chosen as the L9 (34)experimental factor. The optimal condition was 10.6% K3PO4, 42.5% methanol, 5% Ganoderma lucidum extractive, the pH of the lower phase was adjusted to 4.0 after removing the upper phase. Under these conditions, (10.13±0.19) mg/g Ganoderma lucidum triterpenoids were obtained, which was 36.5% higher than that prepared by the traditional method (7.42±0.09) mg/g. The methanol/K3PO4 aqueous two-phase system is a simple, efficient method for extraction and purification triterpenoids from Ganoderma lucidum.
