聚乙二醇-硫酸铵双水相体系萃取松花粉中总黄酮的工艺研究

Study on the extraction of total flavonoids from Pine Pollen in PEG400/ (NH4) _2SO4 aqueous two-phase system

  • 摘要: 采用聚乙二醇-硫酸铵双水相体系萃取纯化松花粉中的总黄酮,确定双水相体系组成为26%PEG400-20%(NH4)2SO4,以萃取率和分配系数为指标,考察p H、温度和Na Cl添加量对双水相萃取分配行为的影响,并结合正交实验优化萃取工艺参数。确定最佳萃取工艺条件为Na Cl添加量为2.0%、p H4、温度为40℃,在此条件下松花粉总黄酮萃取率达到98.93%,其中Na Cl添加量对于萃取分配行为的影响最大。此方法操作简便,水环境温和,分相时间短、萃取率较高,是松花粉总黄酮等天然活性产物分离纯化的一种有效方法。 

     

    Abstract: An aqueous two-phase system of PEG400/ ( NH4) 2SO4was used to extract total flavonoids from Pine Pollen. The composition of the aqueous two-phase system was 26% PEG400-20% ( NH4) 2SO4.Effects of the mass fraction of Na Cl, p H and temperature of aqueous two-phase system on the partition ratio and extraction rate of total flavonoids from Pine Pollen were studied through the single factor experiments. The optimal extraction conditions were obtained by the orthogonal experimental design.The optimum conditions were as follows: the mass fraction of Na Cl at 2%, p H at 4 and temperature at 40 ℃, and the extraction rate can reach up to 98.93%.The Na Cl addition had the most effect on flavonoids partition.The aqueous two-phase extraction has the advantages of simple and convenient procedure, moderate aqueous environment, shorter extraction time and higher extraction rate, which is effective for the separation and purification of total flavonoids.
