超声-微波协同提取蝉花虫草腺苷条件优化及不同产地腺苷测定

Determination and optimal extracted conditions of adenosine in Isaria cicadae mycelium from different areas by ultrasonic-microwave synergistic extraction method

  • 摘要: 以野生蝉花虫草子实体为材料,采用超声-微波协同法提取蝉花虫草腺苷并对其提取条件进行优化,经单因素及正交实验,确定优化的提取条件:微波功率150 W、超声-微波协同提取时间120 s、料液比为1∶30、甲醇体积分数为20%,蝉花虫草腺苷提取率为(0.53±0.02)mg/g;并利用高效液相色谱法测定不同产地蝉花虫草菌丝体腺苷含量,结果表明:测得4个产地的蝉花虫草菌丝体腺苷含量为0.140.40 mg/g,其中井冈山分离获得JJS0017菌株的菌丝体腺苷含量最高(0.40 mg/g),其次为九江市莲花镇莲花林场分离获得JJS0007菌株,江西省鄱阳县饶埠镇分离获得JJS0032菌株的菌丝体腺苷最低(0.14 mg/g)。本文可为蝉花虫草的开发利用提供一定的理论依据。 

     

    Abstract: Taking the fruiting bodies of Isaria cicadae as materials, ultrasonic-microwave synergistic extraction method was used to extract adenosine in I. cicadae mycelium and the extraction conditions were optimized. According to the single factor and orthogonal experiments, the optimal extraction conditions were 150 W of microwave power, 120 s of ultrasonic microwave synergistic extraction time, 1 ∶ 30 of solid-liquid ratio and 20% of methanol volume fraction. The content of adenosine in mycelium of I.cicadae determined was ( 0.53 ± 0.02) mg/g.The adenosine contents in different areas were determined by high performance liquid chromatography.The results showed that the adenosine contents of I. cicadae provided from four areas were ranged from 0.14 to 0.40 mg/g. The content of mycelium adenosine in the strain JJS0017 isolated from Jinggangshan was highest, which was followed by the strain JJS0007 obtained from Lianhua town, Jiujiang city.The lowest content was 0.14 mg/g, which was from the strain JJS0032 from Rao town, Poyang county, Jiangxi province.This article could provide a theoretical basis for the development and utilization of I.cicadae.
