响应面法优化止泻木子总生物碱的提取工艺

Optimization of extraction process for total alkaloids from Holarrhena antidysenteriaca by response surface methodology

  • 摘要: 本文对止泻木种子中总生物碱的提取工艺进行优化。在单因素实验的基础上,通过四因素三水平进行响应面设计实验,以总生物碱得率为响应值进行响应面分析,优化总生物碱的提取工艺参数。结果表明各因素对止泻木子中总生物碱提取得率的影响顺序依次为:提取温度>乙醇浓度>提取时间>料液比。止泻木子中生物碱的最佳提取工艺为乙醇浓度85%、提取时间3 h、料液比1∶10、提取温度80℃。此条件下,止泻木子中总生物碱的含量为(23.8±0.12)mg/g。此方法可靠,提取率高,适于止泻木中的总生物碱提取。 

     

    Abstract: The extraction of total alkaloids from Holarrhena antidysenteriaca was optimized.Single factor experiment was employed to identify four main process parameters.The selected variables were optimized by response surface analysis based on the extraction yield of total alkaloids.The results showed that the effect of the investigated factors on the total alkaloids extraction yield was in the order as: extraction temperature > ethanol concentration > extraction time > matedal-to-solven ratio.The optimal extraction parameters were 85% ethanol and subsequent extraction at 80 ℃ for 3 h with a matedal-to-solven ratio1∶ 10 g/m L. The extraction yield of total alkaloids obtained was ( 23.8 ± 0.12) mg/g under these conditions.This described method was reliable.Highly extraction and suitable for the extraction of total alkaloids from Holarrhena antidysenteriaca.
