Abstract:
To explore the optimum extraction conditions of squid ink polysaccharides from Sepia pharaonis squid ink, this study was conducted to provide a theoretical basis for the development and utilization of squid ink. By single factor tests, enzyme dosage,solid- liquid ratio, reaction temperature, reaction duration and p H were investigated. And on the basis of single factor tests, according to the( Box- Behnken) experimental design protocol, enzyme dosage,solid- liquid- ratio, reaction temperature, and p H were selected to the four factors and the response surface experiment was carried out by response surface analysis. The results showed that their optimal conditions were defined as follows: enzyme amount 46 mg / g,solid- liquid- ratio 1∶ 19.7,reaction temperature 54.1 ℃and p H9.0. Under this condition, the polysaccharide yield was as high as 2.140%.
