聚乙烯吡咯烷酮/K2HPO4双水相体系分配α-乳白蛋白和β-乳球蛋白的研究

摘要: 本文研究了聚乙烯吡咯烷酮PVP/K2HPO4双水相技术分配α-乳白蛋白和β-乳球蛋白的工艺,考察了PVP分子质量、PVP浓度、K2HPO4浓度、p H、温度等不同因素对双水相成相行为和蛋白质分配行为的影响。研究结果表明:分配α-乳白蛋白和β-乳球蛋白的最佳工艺:30℃,分子量10000 u的PVP浓度为16%(w/w),K2HPO4浓度为14%(w/w),p H为7.5。在此条件下,α-乳白蛋白和β-乳球蛋白分别分配在双水相中的上相和下相,其分配系数分别为7.32和0.38,回收率为88.29%和74.07%。Abstract: In the present study,the partitioning of α- LA and β- LG in aqueous two- phase system of polyvinylpyrrolidone- potassium phosphate was investigated.The partitioning of proteins in this system depends on PVP molecular weight,the polymer,salt weight percents,p H and temperature was used to study the effects of different parameters on partitioning of α- LA and β- LG.The optimum conditions of distribution of α- LA and β- LG were 16%( w / w) and 14%( w / w) of the amount of PVP weight percent 10000 u and K_2HPO_4 separately,temperature for 30 ℃,and p H7.5.Results indicated that α- LA and β- LG were assigned in the phase of the upper and the bottom of the aqueous two- phase system respectively.The corresponding partition coefficients were 7.32 and 0.38,and the rate of partition were 88.29% and 74.07% according to the partition of α- LA and β- LG.
