核壳型色谱柱-反相高效液相色谱-蒸发光散射检测法同时测定饮料中5种人工合成甜味剂

Simultaneous determination of five intense sweeteners in beverages by core- shell column- HPLC- ELSD

  • 摘要: 建立了一种同时测定饮料中主要限用人工合成甜味剂:安赛蜜、糖精钠、甜蜜素、三氯蔗糖和阿斯巴甜的反相高效液相色谱-蒸发光散射检测法。采用核壳型HALO色谱柱,甲醇/甲酸-三乙胺缓冲溶液(p H=4.5)为流动相,梯度洗脱,ELSD检测,结果表明5种甜味剂在4.5 min内能够实现完全分离。线性范围为5μg/m L500μg/m L,线性相关性系数大于0.997,方法最低检出限为2μg/m L,定量限为5μg/m L。在3个添加水平下,样品的加标回收率为83.0%106.7%,相对标准偏差为0.84%3.37%。该方法方便、快速、适用性强并可快速推广,可实现饮料中多种人工合成甜味剂的同时分离检测,同时也适用于无紫外吸收的甜味剂的检测。 

     

    Abstract: An analytical method has been established to detect 5 intense sweeteners( Acesulfame- k,Sodium saccharin,Cyclamate,Sucralose,and Aspartame) simultaneously in beverages by reverse phase high performance liquid chromatography( RP- HPLC). The separation process was carried out on a HALO column with methanol /formic acid—triethylamine buffer solution( p H = 4.5) as mobile phase with gradient elution. The quanlification and quantitation were accomplished by the evaporative light scattering detector. The 5 sweeteners were baseline separated in 4.5 minutes.The linear range was from 5 μg / m L to 500 μg / m L,with correlation coefficient greater than0.997.The LOD was 2 μg / m L and LOQ was 5 μg / m L.An average recovery was between 83.0% to 106.7% and the RSD was 0.84% ~ 3.37%. The results indicated that the method was simple,quick,serviceable and applicable to detection of sweeteners in beverage,it was also suitable for the detection of no ultraviolet absorption of sweeteners.
