UPC~2快速测定葡萄酒中抗坏血酸和异抗坏血酸含量研究

Research method of rapid determination of isoascorbic acid and ascorbic acid in wine by Ultra-performance Convergence Chromatography

  • 摘要: 建立了超高效合相色谱法(Ultra Performance Convergence Chromatography,UPC2)分离和测定葡萄酒中抗坏血酸和异抗坏血酸的方法。超高效合相色谱(UPC2)技术集合超临界流体色谱(Supercritical Fluid Chromatography,SFC)和超高效液相色谱(Ultra Performance Liquid Chromatography,UPLCTM)的技术优点,流动相CO2为主体,0.1%磷酸甲醇为助溶剂。选用Waters BEH色谱柱,流速1.5 m L/min,检测波长为245 nm,方法检出限为2.0 mg/kg,定量限为6.0 mg/kg,线性范围为2.5~80.0 mg/L;加标回收率范围为93.2%~105.9%;相对标准偏差RSD为4.5%~9.8%,该方法操作便捷、检测速度快、准确性高、重复性好、节约成本,能够满足葡萄酒中抗坏血酸和异抗坏血酸的检测。 

     

    Abstract: A new method was developed for the determination of isoascorbic acid and ascorbic acid in wine by Ultra Performance Convergence Chromatography(UPC2). The mobile phase was mixture with supercritical CO2 and methanol(added 0.1% H3PO4) at a flow rate of 1.5 m L/min. Using the Waters BEH column,UV detector was set at a wavelength of 245 nm. The limit of detection was 2.0 mg/kg,the calibration linear range was 2.5~80.0 mg/L,its spiked recoveries were 93.2% ~105.9%,and the relative standard deviation range from 4.5% to9.8%. The method with high efficient,rapid,simplicity,high sensitivity and accurate results,it was applicable for determination of isoascorbic acid and ascorbic acid in wine.
