羊栖菜硫酸多糖的超声辅助提取工艺优化及抗氧化活性研究

Optimization of extraction and antioxidative activity in vitro of sulfated polysaccharides from Sargassum fusiforme (Hary) Setch.

  • 摘要: 为开发利用羊栖菜资源,对羊栖菜多糖(Sargassum fusiforme polysaccharides,SFPS)的提取工艺及其抗氧化活性进行了研究。在单因素实验基础上,通过二次回归正交旋转组合设计对超声波辅助提取SFPS粗品的工艺条件进行优化,得到最佳工艺参数为:超声时间13min、提取温度87℃和液料比46∶1m L·g-1,此时SFPS粗品提取率为8.23%±0.01%。粗品中SFPS含量为31.20%±0.74%,硫酸根含量为10.93%±0.24%;SFPS的硫酸基含量41.88%±2.24%,SFPS为硫酸多糖。在一定范围内,SFPS粗品的抗氧化能力与多糖质量浓度呈线性正相关,对·OH、DPPH·、ABTS+·和O2-·的半数清除浓度(IC50)值分别为0.7068、0.4814、0.2527、1.2350mg·m L-1。 

     

    Abstract: In order to exploit resources of Sargassum fusiforme (Hary) Setch., the extraction and antioxidative activity of the polysaccharides from S. fusiforme ( SFPS) was investigated respectively. Based on single experiments, with yield of crude SFPS as index, extraction temperature, ultrasonic-irradiation time and ratio of liquid to solid were selected as factors, the extraction technology of the crude SFPS was optimized by quadratic regression rotatable orthogonal combinition design method. The results showed that the optimal extraction conditions were 87℃, 46∶1m L·g-1with 13 min. The corresponding extraction yield of crude SFPS was8.23%±0.01%. The sulfate-radical concentration of the crude SFPS was 10.93% ±0.24%. The SFPS content of the crude SFPS was 31.20% ±0.74%. The sulfate-radical concentration of the SFPS was 41.88% ±2.24%. The SFPS were sulfated. There was a linear positive correlation between the antioxidant ability of SFPS and the concentration in certain extent. The half-scavenging concentration IC50 values of the crude SFPS against ·OH, DPPH·, ABTS+· and O2-· radicals were 0.7068, 0.4814, 0.2527, 1.2350mg·m L-1respectively.
