鹿茸和大豆蛋白酶解物3ku超滤组分的制备及其对小鼠脾细胞增殖影响的研究
Preparation of 3ku ultrafiltration fractions from velvet antler and soybean protein hydrolysates and their effect on murine splenocytes proliferation

摘要: 以鹿茸水溶性蛋白和大豆分离蛋白为原料,采用模拟胃肠消化和碱性蛋白酶水解,根据游离氨基含量变化优化水解时间,制备鹿茸蛋白和大豆分离蛋白酶解物。通过超滤分离获得各酶解物分子量<3ku的组分:鹿茸蛋白模拟胃肠消化物组分(VAWP-SGD)、鹿茸蛋白碱性蛋白酶解物组分(VAWP-APH)、大豆分离蛋白模拟胃肠消化物组分(SPI-SGD)、大豆分离蛋白碱性蛋白酶解物组分(SPI-APH),探究各组分对小鼠脾淋巴细胞增殖的影响。结果表明:模拟胃肠消化时间为胃蛋白酶2h+胰酶5h;碱性蛋白酶水解时间4h。在无诱导剂时,VAWP-SGD和SPI-SGD对小鼠脾细胞增殖均有显著促进作用(≥100μg/m L)。VAWP-SGD对Con A诱导的小鼠脾T细胞增殖促进作用最显著,其次是VAWP-APH,SPI-APH无明显作用,SPI-SGD对脾细胞增殖有轻微抑制作用。VAWP-SGD对LPS诱导的小鼠脾B细胞增殖促进作用最强,其次是VAWP-APH,SPI-APH及SPI-SGD。综上,VAWP-SGD对小鼠脾细胞增殖影响最大,对进一步开发鹿茸产品和免疫活性肽具有重要意义。Abstract: Velvet antler water- soluble protein ( VAWP) and soybean protein isolate ( SPI) were hydrolyzed by in vitro simulated gastrointestinal digestion and Alcalase to obtain different protein hydrolysates. The hydrolysis time was optimized according to the changes in the content of α- NH2. The < 3ku fractions of velvet antler water- soluble protein simulated gastrointestinal digest ( VAWP- SGD) , velvet antler water- soluble protein Alcalase hydrolysate ( VAWP-APH) , soybean protein isolate simulated gastrointestinal digest ( SPI- SGD) and soybean protein isolate Alcalase hydrolysate ( SPI- APH) were fractionated by ultrafiltration.The effect of < 3ku fractions on the proliferation of murine splenocytes were studied.Results indicated that the hydrolysis time of in vitro simulated gastrointestinal digestion were 2h with pepsin and 5h with pancreatin. The hydrolysis time by Alcalase was 4h. VAWP- SGD and SPI- SGD significantly promoted the proliferation of resting murine splenocytes ( ≥100μg / m L) .VAWP- SGD showed the highest stimulated effect on Con A- stimulated murine splenocytes proliferation, followed by VAWP- APH. SPIAPH had no obvious effect on Con A- stimulated murine splenocytes proliferation, whereas SPI- SGD inhibited the proliferation of Con A- stimulated murine splenocytes.VAWP- SGD also had the strongest stimulated effect on LPSstimulated murine splenocytes proliferation, followed by VAWP- APH, SPI- APH, and then was SPI- SGD.The above results showed that VAWP- SGD had the strongest stimulated effect on the proliferation of murine splenocytes, which suggested that VAWP- SGD was of great significance for the development of antler products and immune active peptide.
