杆菌肽高产菌株的诱变选育

Mutation breeding of high- yield strain of bacitracin

  • 摘要: 为了选择有效的方法选育出高产杆菌肽的菌种。利用地衣芽孢杆菌FUN-BL为出发菌株,依次进行紫外线诱变和硫酸二乙酯诱变,再通过平板活性筛选法作为初筛和HPLC作为复筛,筛选出效价较高的菌株。出发菌株F0经过紫外线诱变(15W,照射距离30cm,照射时间120s)后,筛选得到高产菌株F167,效价提高了11.76%。菌株F167经过硫酸二乙酯诱变(处理浓度为2%,处理时间为90min)后,筛选得到高产菌株F2141效价提高了32.19%。将高产菌株F2141进行连续5代遗传稳定性实验,效价依次是1003.23、989.34、976.35、960.32、949.72U/m L。通过紫外线诱变和硫酸二乙酯诱变能够选育出具有遗传稳定性的高产杆菌肽的菌株。 

     

    Abstract: The purpose of this article was to select effective method to breed high- yield strains of bacitracin.Using bacillus licheniformis FUN- BL as an aboriginal strains, the positive mutants were screened by cup- plate method and HPLC after ultraviolet and diethyl sulfate. The aboriginal strains F0 mutated through ultraviolet ( 15 W, the irradiation distance of 30 cm and the irradiation time of 120 sec) were screened in repeat to obtain the high- yield strain of bacitracin F167, whose titer was improved by 11.76%.Then F167 mutated through diethyl sulfate ( dealt with2% diethyl sulfate for 90min) were screened in repeat to obtain the high- yield strain of bacitracin F2141, whose titer was improved by 32.19%. Making the high- yield strains F2141 do the genetic stability test for five successive generations, the titer followed 1003.23, 989.34, 976.35, 960.32, 949.72 U / m L. High- yield strains of bacitracin can be bred by ultraviolet and diethyl sulfate.
