交替糖蔗糖酶受体反应产低聚四糖及其条件优化

摘要: 为探究交替糖蔗糖酶这一新型葡聚糖蔗糖酶产低聚糖的受体反应,确定其产低聚四糖的最优条件。采用嗜柠檬酸明串珠菌SK 24.002产交替糖蔗糖酶,以蔗糖和麦芽糖为底物,利用该酶的受体反应产低聚四糖,并对该反应进行了条件优化。结果表明,最优反应条件为:100g/L蔗糖+100g/L麦芽糖+0.3U/m L酶液,反应时间为5h,反应温度为40℃,反应体系为50mmol/L p H5.4的醋酸缓冲液,在此条件下,低聚四糖的产量最高可达52.8g/L。通过控制受体反应的条件,可有效的提高产物低聚糖的产量。Abstract: The object of the study was to determine the optimal conditions of receptor reaction catalyzed by alternansucrase, a new kind of glucansucrases, to produce oligosaccharide DP4.Leuconostoc corium SK24.002 was used as the culture to produce alternansucrase, and receptor reaction of alternansucrase was conducted using sucrose and maltose as substrates. Results showed that the optimal conditions of receptor reaction catalyzed by alternansucrase were: 100 g / L sucrose + 100 g / L maltose + 0.3U / m L enzyme solution in 40℃ in 50 mmol / L p H5.4acetate buffer for 5h.Under these conditions, the maximum yield of oligosaccharide DP4 finally achieved 52.8g / L.The products of receptor reaction catalyzed by alternansucrase can be greatly impacted by the reaction conditions.
