云南牛肝菌的内生真菌分离、鉴定和ITS序列特征研究

Isolation and identification of endophytic fungi from different Boletus in Yunnan and ITS region sequence analysis

  • 摘要: 以云南产4种牛肝菌:小美牛肝菌Boletus speciosus Frost、铜色牛肝菌Boletus aereus、黄皮疣柄牛肝菌Leccinum crocipodium、美味牛肝菌(白牛肝)Boletus edulis新鲜子实体为材料,经无菌技术分离子实体内部组织培养,从中获得5株内生真菌,其中YX1-6分离于黄皮疣柄牛肝菌、YX1-7分离于美味牛肝菌、QJ3-6分离于铜色牛肝菌、QJ3-7分离于美味牛肝菌、QJ3-8分离于华丽牛肝菌。根据菌株的形态特征和ITS序列分析对5株菌进行鉴定,结果表明:YX1-6为毛栓菌(Trametes hirsuta),YX1-7为金色毛壳菌(Chaetomium aureum),QJ3-6为假丝酵母属(Candida sp.),QJ3-7为被孢霉属(Mortierella sp.),QJ3-8为散囊菌属真菌(Eurotium sp.)。经ITS rRNA二级结构辅助分析可看出,所获的牛肝菌内生真菌差异较大,揭示了云南野生牛肝菌内生真菌的多样性特征。 

     

    Abstract: Five strains of fungal endophytes, named YX1-6, YX1-7, QJ3-6, QJ3-7, QJ3-8, were isolated from four spieces of Boletus specious Frost, Boletus aereus, Leccinum crocipodium, Boletus edulis which were collected in Yuxi and Qujing areas of Yunnan Province.The morphyological characters of the strains were observed, and their ITS sequences were amplified and sequenced.Phylogenetic tree was constructed by the obtained molecular data.The results showed that YX1-6 was Trametes hirsuta., YX1-7 was Chaetomium aureum., QJ3-6 was Candida sp., QJ3-7 was Mortierella sp., QJ3- 8 was Eurotium sp.. The secondary structure of ITS rRNA in these five strains predicted with the softwares of DNA STAR, RNAVIZ2.0. and MEGA5.1 indicated that fungal endophytes from Boletaceae were different and diversified.
