酪蛋白磷酸肽副产物中ACEI肽的富集工艺研究

Enrichment process of angiotensin I-converting enzyme inhibitory peptides from casein peptide phosphate by-products

  • 摘要: 以工厂生产酪蛋白磷酸肽后的副产物(CPPBP)为原料,在研究其分离特性的基础上,初步去杂,并借助马尿酸法和HPLC法定位活性最高多肽的位置。接着,以高活力ACEI肽得率为指标,设计三因素三水平的正交实验优化富集工艺,最终研发一种二段量化富集ACEI肽的新工艺。该工艺第一段以CPPBP饱和多肽液为原料,设定沉淀条件为pH2、-16℃、沉淀1h,得到粗富集物(P粗)。第二段再以P粗饱和液为原料,在pH4、乙醇浓度70%、-20℃条件下,沉淀1h。最后产物得率为19.11%,经活性测定,ACE抑制率为46.29%。 

     

    Abstract: In this paper, a novel two-stage enrichment process was developed for mass-producing ACE-inhibitory peptides by using casein peptide phosphate production by- products ( CPPBP) .After pre- concentrating active peptides on the basis of analyzing separation properties of CPPBP, the high efficient components were located by hippuric acid method and HPLC method.And then a three factor and three levels orthogonal experiment, taking yield of high efficient components as index, was designed to optimize the process conditions of concentrating ACE- inhibitory peptides.In first stage, saturated CPPBP solution was used as material and precipitation conditions were set as pH2 and-16℃ for 1h.The pre- concentration was named as Pcrude.In second stage, the final product was precipitated from saturated Pcrudeunder pH4, Ethanol concentration 70% and- 20℃ for 1h.The yield of enrichment process was 19.11% with an inhibition of 46.29% to ACE.
