倒卵叶五加中总皂苷的纯化工艺研究

Study on purification of total saponins from Acanthopanx obouatus Hoo

  • 摘要: 目的:利用大孔吸附树脂纯化技术研究倒卵叶五加中总皂苷的最优纯化工艺。方法:通过对NKA-9、D101、LS-303、LS-45和LS-21等5种大孔吸附树脂的静态实验,筛选得到最佳树脂;在此基础上,通过华体会(中国)实验确定最佳树脂对倒卵叶五加总皂苷的纯化的最优工艺参数。结果:LS-303树脂纯化倒卵叶五加总皂苷的效果最好,最优工艺条件为:上样液浓度约为3mg·mL-1,上样量为9BV,上样流速为2BV/h;解吸剂为80%的乙醇,解吸流速2BV/h,洗脱剂用量为2.5BV。结论:LS-303型大孔吸附树脂可有效分离纯化倒卵叶五加中的总皂苷。 

     

    Abstract: Objective:Inquiry into the optimal purification condition of total saponins from Acanthopanx obouatus Hoo with macroporous adsorptive resins. Methods:Screen the optimal macroporous resin by the static test on five different models macroporus resin. Optimize the technological parameter by investigating the absorption and desorption performances of the optimal macroporous resin on total saponins from Acanthopanx obouatus Hoo. Results:LS-303 macroporous resin was the optimal macroporous resin on total saponins from Acanthopanx obouatus Hoo. Optimum technology was that:the concentration of sample solution was 3mg/mL with loading amount of 9BV, the velocity of sample loading was 2BV/h, the eluent was 80% alcohol, the elution velocity was2BV/h, the elution volume was 2.5BV. Conclusion:LS-303 macroporous resin in purifying total saponins from Acanthopanx obouatus Hoo and the advantages of the developed method for industrial application, liked simple operation, safety, and low cost.
