酶解酸枣仁蛋白制备抗氧化肽的研究

Preparation of antioxidant peptides from Zizyphi Spinosi Semen protein by enzymatic hydrolysis

  • 摘要: 为优化Alcalase蛋白酶酶解酸枣仁蛋白制备抗氧化肽的工艺条件,以DPPH自由基清除率作为指标,通过单因素试验考察酶解温度、底物浓度、酶与底物浓度比(E/S)、酶解pH和时间对DPPH自由基清除率的影响,在单因素试验的基础上,采用Box-Behnken设计,建立并分析各因素与清除率之间关系的数学模型;并对酸枣仁蛋白酶解产物的氨基酸组成及含量进行分析。结果表明:酶解酸枣仁蛋白制备抗氧化肽的最佳工艺条件为酶解温度51.2℃、酶解pH8.1、底物浓度4.7%,在此条件下DPPH自由基清除率的最大理论值为91.59%,该回归模型具有较好的预测性能,可用于指导生产实践。酸枣仁蛋白酶解产物共含有17种氨基酸,富含脯氨酸、甘氨酸和谷氨酸。 

     

    Abstract: Optimization enzymolysis technology for production of antioxidant peptides of Zizyphi Spinosi Semen protein were studied. The antioxidant activities of enzymatic hydrolysates from spine date seed protein were investigated by 1, 1- diphenyl- 2- picrylhydrazyl ( DPPH) radical scavenging activities. Effect of enzymolysis temperature, substrate concentration, ratio of enzyme to substrate ( E/S) , enzymolysis pH and hydrolysis time on the DPPH scavenging rate of enzymatic hydrolysate from spine date seed protein were taken by the single factor experiments. Technology parameters were optimized by Box- Behnken statistical design. The amino acid composition and content of enzymatic hydrolysate of Zizyphi Spinosi Semen protein was measured by protein sequencer.The results showed that when the extraction parameters were controlled at enzymolysis temperature 51.2℃, enzymolysis pH8.1, substrate concentration 4.7%, the DPPH scavenging rate could be up to 91.59%. The predictive models are feasible in practice.Enzymatic hydrolysate of Zizyphi Spinosi Semen protein, consisting of 17 kinds of amino acid, was rich in proline, glycine and glutamic acid.
