从制备血红蛋白肽的血渣中提取血红素

Extraction hemin from blood residue of hemoglobin peptide

  • 摘要: 目的:优化酶解血渣中提取血红素的最佳提取条件。方法:取酶解制备血红蛋白肽的血渣为原料,采用酸性丙酮法,用L9(34)正交实验,对影响提取效果的因素(盐酸丙酮比A、料液比B、搅拌时间C)进行优选。结果:血渣中提取血红素的最佳工艺条件是盐酸丙酮比为4∶100、料液比为1∶5、搅拌时间为20min,提取的血红素含量可以达到97%以上,铁含量7.8%以上,提取率可达到62.89%。结论:此工艺易于操作、重复性好、所得产品纯度高,适合工业化生产。 

     

    Abstract: Objective: To optimize the extraction process of hemin in the enzymatic hydrolysis blood residue.Method:Taking the enzymatic hydrolysis blood residue of preparation hemoglobin peptide as raw material, the hydrochloric acid consumption A, solvent consumption B, mixing time C were optimized by the L9 ( 34) orthogonal design.Results: The optimum extration condition were as follows: hydrochloric acid to acetone ratio = 4 ∶ 100, 5 folds of Acidic acetone, stirring 20min. The purity of extracted hemin could reach more than 97%, the iron conten could reach more than 7.8%.Conclusion: This method was easy to operate, had better reproducible and could be used for industrial production.
