产阿魏酸酯酶乳酸菌的乳杆菌筛选以及基因克隆

Screening and gene cloning of ferulic acid esterase by Lactobacillus strains

  • 摘要: 运用平板法和高效液相色谱法筛选出一株具有阿魏酸酯酶活性的发酵乳杆菌(Lactobacillus fermentum ATCC9338),其酶活力为260mU/L,并成功运用鸟枪法对该菌株进行阿魏酸酯酶克隆并测序,为产阿魏酸酯酶益生菌的开发提供理论依据。 

     

    Abstract: The production of feruloyl esterase activity by Lactobacillus could be screened by assaying transparent zone on the selective agar plate and high efficiency liquid chromatography. The results of experiment showed that only Lactobacillus fermentation ATCC 9338 had feruloyl esterase activity, and the enzyme activity was 260mU/L. Shotgun cloning method was used to clone and sequence feruloyl esterase gene from this strain. This study will provide theoretical basis for feruloyl esterase-producing probiotics.
