茶枝柑皮多糖对PC12细胞氧化损伤的保护作用

摘要: 目的:研究茶枝柑皮多糖对H2O2诱导的PC12细胞氧化损伤的保护作用。方法:MTT法检测细胞存活率,建立H2O2诱导PC12细胞氧化损伤模型。比色法测定细胞内丙二醛(MDA)含量、超氧化物歧化酶(SOD)以及谷胱甘肽过氧化物酶(GSH-Px)的活性。结果:100μmol/L H2O2诱导PC12细胞4h,细胞呈现明显损伤形态,细胞内MDA含量升高,SOD和GSH-Px活性降低。茶枝柑皮多糖可明显改善PC12细胞损伤,显著降低细胞MDA含量,极显著提高SOD和GSH-Px活性。结论:茶枝柑皮多糖对H2O2诱导PC12细胞损伤具有明显的保护作用,其作用机制可能与提高PC12细胞的抗氧化酶活性有关。Abstract: Objective:To study the protective effect of polysaccharide from Citrus reticulata ‘Chachi' Peel on H2O2-induced oxidative injury in PC12 cells.Methods:Cell viability was assayed by MTT method, the toxicity model was established by treating PC12 cells with H2O2.The level of MDA, SOD and GSH-Px were measured by spectroscopy, respectively.Results:After being exposed to 100μmol/L H2O2 for 4 hours, typical morphological characteristics of injury were observed.The level of MDA increased significantly, while that of SOD and GSHPx decreased significantly in H2O2- induced group compared with that of the sham group.Polysaccharide from Citrus reticulata ‘Chachi' Peel could decrease MDA content and increase SOD and GSH-Px activity significantly.Conclusions:Polysaccharide from Citrus reticulata ‘Chachi' Peel had protective effect on PC12 cells injury induced by H2O2 in culture.The protective effect might be related to the increased activity of antioxidant enzymes.
