曲霉属蛋白表达中高效启动子的应用研究

  • 摘要: 曲霉属丝状真菌,尤其是黑曲霉(Aspergillus niger)和米曲霉(Aspergillus oryzae),常被作为宿主菌表达生产各类蛋白并在工业上得以广泛使用。工业生产中,为了提高蛋白产量,获取更大的经济效益,需要对影响蛋白表达的各种因素进行改进,使用高效的启动子便是其中策略之一,强启动子的使用能够从转录水平上提高同源或是异源蛋白在曲霉中的表达产量。该文介绍了曲霉蛋白表达过程中常见启动子的使用情况,重点阐述了国内外改进启动子活性构建高效启动子的几种策略,对新型启动子在曲霉蛋白表达中的应用做出了展望。 

     

    Abstract: Aspergillus species, especially Aspergillus niger and Aspergillus oryzae are widely used as expressed strains to produce a large number of protein products industrially.To increase the industrial production and achieve more economic benefits, it still needs to take measures to improve protein expression.There are many factors that may affect protein production, among which promoter efficiency is the most critical one.As strong promoters could be used to increase both homologous and heterologous protein expression at the transcriptional level, the choice and improvement of promoters in Aspergillus are very important. This review summarized some commonly used promoters in Aspergillus, and had focused on varied strategies to improve promoter efficiency. In addition, the application perspectives of modified promoters in protein expression had also been briefly described.
