Abstract:
The production of succinic acid by bacterial fermentation has a great prospect for industrial application. In this study, the fermentative productions of succinic acid by recombinant Escherichia coli WS100 (△ldhA, △adhE, △pflB, △poxB, △ackA) with different media were investigated. 4L NBS medium and corn steep liquor medium were utilized respectively for fed batch fermentations with E.coli WS100 in fermenters. After 83h of fermentation, 96.17g/L glucose was consumed in corn steep liquor medium, with the production of 63.45g/L succinate. The productivity and yield reached 0.76g/ (L·h) and 65.98%, respectively. In NBS medium, 110.30g/L glucose was consumed with the production of 84.98g/L succinate. The productivity and yield reached 1.02g/ (L·h) and 77.04%, respectively. The results indicated fine succinate fermentative production was by WS100 in both media, and with higher productive amount, productivity and yield in NBS medium.
