响应面法优化红雪茶中类胡萝卜素的提取工艺
Optimization of extraction conditions of carotenoids from Lethariella spp. by response surface methodology

摘要: 为确定红雪茶类胡萝卜素的最佳提取工艺条件,在单因素实验的基础上,选择丙酮为浸提溶剂,料液比、提取温度、提取时间作为自变量,以类胡萝卜素特定波长下吸光度值作为响应值,基于Box-Behnken中心组合方法进行三因素三水平的实验设计,根据响应面结果,建立了回归模型。研究结果表明,红雪茶类胡萝卜素的最优提取工艺条件参数为:料液比1∶3(g/mL),提取温度52℃,提取时间20min。在该条件下,红雪茶类胡萝卜素提取液的吸光值为0.3682,实际获取红雪茶类胡萝卜素量22.092μg/g,与预测值相对误差较小。说明该模型拟合度好,优化后得到的提取条件准确、可靠。Abstract: Based on Box-Behnken center composite design within Design-Expert7. 0 software and response surface results, the optimal extraction process for carotenoids from Lethariella spp. were determined and a regression model was established. On the basis of single factor tests, using absorbance values in a specific wavelength of carotenoids as response value, three key factors (solid-liquid ratio, extraction temperature and time) were selected as independent variables and acetone as extraction solvent. The results showed that the optimal extraction process of carotenoids from Lethariella spp. was obtained as followed:solid-liquid ratio of 1∶3 (g/mL) , extraction temperature of 52℃, and extraction time of 20min. Under these conditions, the extraction of carotenoids from Lethariella spp. absorbance was 0.3682, i. e. 22.092μg from per g Lethariella spp. raw materials. The results showed that regression equation fit well with experimental data because of a good accordance between the predicted and observe values. Meanwhile the optimized technology was stable and viable with minor error and good degree.
