效应面法优选碎米制备注射用葡萄糖中糖化液离子交换工艺的初步研究

Preliminary optimal ion-exchange technology of saccharification liquid made from broken rice for preparing glucose injection by response surface methodology

  • 摘要: 采用星点设计-效应面法优化碎米糖化液离子交换的工艺条件。以树脂装量、pH和洗脱流速为考察因素,以洗脱指数为效应值,分别采用多元线性回归和二项式方程拟合,用效应面法优化工艺,并进行预测分析。碎米糖化液离子交换的优化工艺为:在两支10mm×20cm的层析柱中分别加入阳、阴离子交换树脂10g,分别调pH为4.0、10.0后,在常温下以10d/min的速度洗脱。本文所建立的离子交换工艺简单可行,可为进一步制备注射用葡萄糖的离子交换工艺研究提供实验依据。 

     

    Abstract: To optimize the ion-exchange technology conditions of saccharification liquid from broken rice by central composite design and response surface methodology. Taking resin loading quantity, pH and elution speed as investigation factors, and the elution efficiency as effect size.The multiple linear regression and binomial equation were fitted respectively, and response surface methodology was adopted to optimize the technology, predict and analyze the possible result. The optimum technology for ion- exchange conditions of saccharification liquid from broken rice was like this:10g cation exchange resin ( pH 4.0) and 10g anion exchange resin ( pH 10.0) were added into two chromatographic columns ( 10mm × 20cm ) severally, eluted at the speed of 10d /min under room temperature.The above-mentioned experiment proved the ion-exchange technology was simple, and practicable, and it could provide experimental basis for ion-exchange technology research on further preparation of the glucose injection.
