响应面法优化枇杷核多糖的提取工艺

Optimization of extraction conditions of polysaccharide from loquat core by response surface methodology

  • 摘要: 采用响应面分析法(RSM)优化超声波辅助酶法提取枇杷核多糖的工艺条件,在单因素实验基础上,选取纤维素酶用量、酶解pH、酶解温度、酶解时间为影响因子,以枇杷核多糖得率为响应值,应用Box-Behnken中心组合实验设计建立数学模型,进行响应面分析,得出优化后的提取工艺条件为:料液比1:15,超声处理温度60℃,超声处理时间30min,纤维素酶用量1.8%,酶解温度49℃,酶解时间1.8h和酶解pH4.7。在此优化工艺条件下,干燥处理后枇杷核多糖的得率为9.96%。 

     

    Abstract: Response surface methodology (RSM) was applied to optimize the extraction conditions of loquat core polysaccharides.Based on single factor experiment, the amount of cellulase, pH, enzymolysis temperature and reaction time were chosen as influencing factors, the loquat core polysaccharides yield was selected as response value.The mathematical model was established by Box-Behnken central composite design.The results showed that the optimal extraction conditions were as follows:material-water ratio 1:15, ultrasonic temperature 60℃, ultrasonic time 30min, the amount of cellulase 1.8%, enzymolysis temperature 49℃, enzymolysis time 1.8h and pH4.7.In this optimal process condition, the yield of loquat core polysaccharides was 9.96%.
