发酵液中γ-聚谷氨酸含量快速测定方法研究

摘要: 通过γ-聚谷氨酸(γ-PGA)溶液与十六烷基三甲基溴化铵(CTAB)的氢氧化钠溶液反应形成混悬液,配制不同浓度的γ-PGA标准品溶液,与CTAB溶液形成反应体系并进行全波长扫描,绘制不同波长下标准曲线,并对CTAB试剂浓度、反应时间和温度对反应体系的影响分别进行了比较优化。研究结果为:CTAB比浊法最佳检测波长为250nm,CTAB溶液质量浓度为5g/L,反应温度为环境温度,络合时间为3min,回归线性方程y=0.0335x-0.1519,R2=0.9995,发酵液中加标平均回收率为106.6%,平均RSD值为2.56%。应用比浊法测定γ-PGA的含量快速、简洁、重现性好,可用于发酵液中γ-PGA浓度的检测。Abstract: A turbid solution was formed through the reaction between γ-PGA and Cetyl Trimethyl Ammonium Bromide (CTAB) sodium hydroxide solution, the preparation of different concentrations of γ-PGA standard solution with CTAB solution to form a reaction system, and the formation to make a full wavelength scan, draw the standard curve of different wavelengths, and CTAB reagent concentration, reaction time and temperature on the reaction system were optimized.The results showed that CTAB turbidimetry best detection wavelength was 250nm, CTAB mass concentration of 5g/L, reaction temperature was room temperature, complexing 3min.The linear regression equation y=0.0335x-0.1519, R2=0.9995.The precision and recovery of this method were within 2.56% and 106.6%.The turbidimetric method has advantages of convenience, simplicity and good repeatability and can be used to detect the γ-PGA concentration in the fermentation broth.
