槐花中芦丁超声辅助提取工艺优化及抗氧化活性研究

摘要: 用超声辅助醇提工艺,进行正交实验设计,优化出从槐花中提取芦丁的最佳条件,通过测定自由基清除率确定了芦丁的抗氧化活性,最后将其应用到提高辣椒红色素稳定性实验中,并与几种常用的抗氧化剂性能进行比较。结果表明,醇提工艺的最佳参数为:乙醇浓度60%、料液比(g/mL)1:10、提取温度45℃,超声时间20min,在此条件下,得率可达6.10%;芦丁浓度与自由基清除率均存在良好的线性关系,其中清除DPPH自由基和超氧阴离子自由基的能力较强,其IC50分别为28.39、69.88μg/mL;在用于提高辣椒红色素的稳定性实验时,芦丁作用仅次于VE,而强于VC和BHT。Abstract: The optimum condition for rutin from sophora japonica of extraction assisted with ultrasonic wave was studied by the orthogonal design mode.Its antioxidant activity was analysed using super oxide anion radical method.The result showed that the optimal extracting conditions were as follows:extracion solvent 60% ethanol, ratio of solvent to solute (v/w) 10, extracion temperature 45℃, and extraction time 30min.There were good relationship between contents of rutin and activities of scavenging hydroxyl free radical, superoxide anion free radical and DPPH radical and appeared linear relationships.The abilities of scavenging DPPH radical and superoxide anion free radical were stronger, the IC50 were 28.39μg/mL and 69.88μg/mL, respectively.When adding the antioxidant, stability of red peppers haematochrome was improved.Compared with VC, VE and BHT, rutin was superior to VE and BHT and next to VE.
