大孔吸附树脂纯化枸杞总黄酮的研究

Separation and purification of total flavonoids in Lycium barbarum L.from Qaidam Basin by macroporous resin

  • 摘要: 研究大孔吸附树脂纯化柴达木枸杞总黄酮的工艺条件及参数。通过比较11种大孔吸附树脂的静态吸附解吸性能,筛选出适合纯化柴达木枸杞总黄酮的树脂类型;并进行华体会(中国)吸附解吸实验,利用单因素和响应面法优化大孔吸附树脂纯化柴达木枸杞总黄酮的工艺条件。结果表明,HPD400型大孔吸附树脂的纯化效果最佳,最佳纯化工艺条件:以16.0mLpH为4.0的柴达木枸杞总黄酮粗提液上柱,流速1.0mL/min;充分吸附后用3BV去离子水洗柱,然后用23.0mL80%乙醇溶液以流速1.0mL/min进行解吸。此工艺的平均回收率为89.92%;经HPD400树脂纯化后提取物中总黄酮含量从10.80%提高到27.62%。 

     

    Abstract: To optimize the purification technology and parameters of macroporous resins to total flavonoids in Lycium barbarum L.from Qaidam Basin.The HPD400 was optimized from 11 types of macroporous resins by comparing their static adsorption and desorption performances with the total flavonoids in Lycium barbarum L.from Qaidam Basin.Single-factor tests and response surface method (RSM) were used for optimizing the purification technology.The optimal purification condition as follows:sample loading amount were 16.0mL, pH4.0, and the velocity of sample solution 1.0mL/min, rinsing the column with 3BV deionized water after completely adsorption, and then 80% ethanol as eluent, the volume 23.0mL and the velocity 1.0mL/min.Under these optimal parameters, total flavonoids recovery rate were up to 89.92%, and the content of total flavonoids purified by HPD400 macroporous resin increased from 10.80% to 27.62%.
